چکیده

Multipotent, self-renewing neural stem cells reside in the embryonic mouse telencephalic germinal zone. Using an in vitro neurosphere assay for neural stem cell proliferation, we demonstrate that FGF-responsive neural stem cells are present as early as E8.5 in the anterior neural plate, but EGF-responsive neural stem cells emerge later in development in a temporally and spatially specific manner. By separately blocking EGF and FGF2 signaling, we also show that EGF alone and FGF2 alone can independently elicit neural stem cell proliferation and at relatively high cell densities separate cell nonautonomous effects can substantially enhance the mitogen-induced proliferation. At lower cell densities, neural stem cell proliferation is additive in the presence of EGF and FGF2 combined, revealing two different stem cell populations. However, both FGF-responsive and EGF-responsive neural stem cells retain their self-renewal and multilineage potential, regardless of growth factor conditions. These results support a model in which separate, lineage-related EGF- and FGF-responsive neural stem cells are present in the embryonic telencephalic germinal zone.

Study on Effect of Head, Tail, and Limbud extracts of Mouse on Differentiation of Hair Follicle Stem Cells to Neural cells

Introduction: Adult stem cells are the group of cells which conserve their nature in tissues and organs among other cells. In recent years, the researchers reported the existence of stem cells on the Bulge of hair follicles near to the smooth muscle. It is possible to differentiate these stem cells to neural cells by induction of Shh, FGF, and RA factors. Because of existence of these factors ...

Cholinergic Differentiation of neural precursor cells derived from mouse embryonic stem cells increased by Shh, LIF and RA

Introduction Cholinergic system is one of the important systems of mammalian CNS. Cholinergic neurons distributed in brain and spinal cord and contributed to principal functions like: consciousness, learning and memory, and motor control. In this study we investigated the differentiation potentiality of mouse embryonic stem cells toward cholinergic neurons. The aim of this study was to evaluate...

The effect of Fibroblast Growth Factor-2(FGF-2) and retinoic acid on differentiation of mouse embryonic stem cells into neural cells

Introduction: Embryonic Stem (ES) cells as pluripotent cells derived from the inner cell mass of blastula can differentiate to neural cells in vitro and this property is valuable in studies of neurogenesis and in the generation of donor cells for transplantation. In this regard, the propose of this research, was the study of the role of two important factors in the development of neural syst...

EGF-responsive neural stem cells are a transient population in the developing mouse spinal cord.

The adult mouse forebrain, which exhibits substantial ongoing cell genesis, contains self-renewing multipotent neural stem cells that respond to epidermal growth factor (EGF), but the adult spinal cord, which exhibits limited cell genesis, does not. Spinal cord development is a process characterized by defined periods of cell histogenesis. Thus, in the present study we asked whether EGF-respons...
